چکیده

The effects of 9-butyl-6-thioguanine (9-BTG)1 on the utilization of preformed purines by the Ehrlich ascites tumor were studied in vivo. The results showed that 9-BTG inhibited the utilization of preformed purines for DNA synthesis under conditions in which UNA synthesis and the incorporation of glycine into proteins and nucleic acids were unaffected. Studies on the incorporation of glycine-2-C14 and adenine-8-C14 into adenine nucleotides suggested that there was partitioning of the acid-soluble adenine into separate pools. The synergistic effect obtained when 9-BTG and azaserine were combined for therapy of the Ehrlich ascites tumor can be explained on the basis that, of the two available sources of purine nucleotides for DNA synthesis, 9-BTG inhibits the utiliza tion of host purines and azaserine blocks de novo purine synthesis.

Formation of 6-thioguanine and 6-mercaptopurine from their 9-alkyl derivatives in mice.

Several 9-alkyl, 6-thiopurines have been reported to have more favorable therapeutic indexes than do the parent drugs, 6-mercaptopurine (MP) and 6-thioguanine (TG). Some of these compounds were reported to be active against cells in culture resistant to 6-thiopurines, and it has been assumed that their mechanisms of action may differ from those of TG and MP. 9-(n-Butyl)-6-thioguanine was essent...

Purinethiols as feedback inhibitors of purine synthesis in ascites tumor cells.

The capacity of several purinethiol derivatives to produce prolongation of survival in mice bearing ascites-cell tumors and to produce feedback inhibition of purine synthesis in vivo in the tumor cells was investigated. Included were tumors susceptible and tumors resistant to treatment with 6-thioguanine. With susceptible tumors, certain of the purinethiols could increase survival of tumor-bear...

The metabolism of 9-butyl-6-thioguanine in normal and neoplastic tissues.

The metabolism of 9-butyl thioguanine (9-BTG)1 was studied in normal tissues and the Ehrlich ascites tumor with and without pretreatment with azaserine. The study of the distribution and metabolism of the drug was facilitated by the synthesis of 9-butyl-6-thioguanine-8-C14. The results showed that 9-BTG was rapidly cleared from the blood, rapidly catabolized, and that the metabolites were rapid...

Quantification of thioguanine-resistant lymphocytes from mice irradiated in vivo.

Adult mice were Co-60 gamma irradiated, and 7 months later splenocytes were isolated, cultured in microwells, and the frequency of hprt-deficient mutants was determined by measuring the cloning efficiency in media with 6-thioguanine. The mutant frequency at 2, 4, and 6 Gy was 1.6 x 10(-5), 4.4 x 10(-5), and 12.7 x 10(-5), respectively. The frequency of spontaneous mutants was 2.5 x 10(-6). The ...
